I thought about this for a moment, pondering various options, and then went "OH WAIT. GLADOS."
If you haven't played the Portal games, you play a woman who is trapped inside an otherwise lifeless lab, being forced to use a portal gun to solve various "tests" by a science obsessed AI called GlaDOS who is by turns politely informative and sadisticly cruel.
She is the best and I love her.
I usually avoid puzzle platformers, but I got Cam to help me through the tougher parts, and loved it. Portal 2 is fantastic too, and fleshes out GlaDOS as a character in a really interesting way.

Ah, and the post asks about a theme song! She has one for each game, both amazing in context: Still Alive and Want You Gone by Jonathon Coulton. EDIT: There's a third, from a Lego Portal game I have never played!
